چکیده

In the previous paper (Jones & Elliott, 1959) it was shown that rat-kidney mitochondria are able to synthesize nicotinuric acid from nicotinic acid and glycine in the presence of fumarate and adenosine triphosphate (ATP). These experiments showed that nicotinuric acid synthesis is dependent upon a suitable source of energy, but did not give any other indications of the mechanism of the reaction. Although the mechanism of nicotinuric acid synthesis has not previously been investigated, the formation of other acyl-glycine compounds, particularly hippuric acid and p-aminohippuric acid, has been extensively studied. The work of Chantrenne (1951) with extracts of acetone-dried ratliver powder first showed that coenzyme A (CoA) was required for hippuric acid synthesis. Subsequently, Schachter & Taggart (1953) showed that hippuric acid synthesis proceeded in two steps, with benzoyl-CoA as an intermediate:

OBJECTIVE Metabolic syndrome is a multiplex disorder and puts patients on the road to type 2 diabetes and atherosclerotic cardiovascular diseases. However, a surrogate biomarker in plasma or urine in fully reflecting features of metabolic syndrome has not been explored. RESEARCH DESIGN AND METHODS Urine metabolomics has potential utility in metabolic profiling because urine metabolites analys...

High-performance liquid-chromatographic determination of free nicotinic acid and its metabolite, nicotinuric acid, in plasma and urine.

We report a liquid-chromatographic procedure for determining free nicotinic acid and a metabolite, nicotinuric acid, in plasma and urine. Five-tenths milliliter of urine or deproteinized plasma is evaporated and the residue analyzed isocratically by reversed-phase ion-pair chromatography, with measurement of the eluted nicotinic acid and nicotinuric acid at 254 nm. Nicotinic acid, nicotinuric a...

The Microbiological Determination of Nicotinic Acid, Nicotinamide, and Nicotinuric Acid

The nicotinic acid assay most generally satisfactory is the Lactobacillus arabinosus procedure (1, 2) as regards sensitivity and non-interference from other compounds. However, this organism responds to nicotinamide and nicotinuric acid as well as to nicotinic acid. Gaines and Stahly (3) have proposed the use of Leuconostoc mesenteroides for the assay of several members of the vitamin B complex...

Studies on the metabolism of nicotinic acid in rabbits.

Several compounds have been described as metabolites of nicotinic acid and nicotinamide in mammals. These are nicotinic acid (I), nicotinamide (2)) nicotinuric acid (2)) trigonelline (2)) W-methylnicotinamide (NMN) (3-6)) and 1-methyl-3-carboxylamide-6-pyridone (7, 8). According to the observations of various workers mammals may be divided into two groups in so far as the metabolism of nicotini...
